I'm going to state one that we all probably know, but in case you are a newbie. A great way to make your podcast ready for iTunes is feedburner.com It's free and its owned by Google (so you don't have to worry about them going out of business.
The great thing is you can put your iTunes specific in formation into it. I like this better than using a plugin (like Podpress) as plugins (like Podpress) may stop to be updated. Then you are stuck.
Feedburner also has some tools to make it easy to promote your podcast, make it easy to subscribe to the podcast, and monetize your podcast as well. AND IT'S FREE
I do not use their download tracking feature. I use other tools (Libsyn, or Podtrac) for that.
